ER stress and apoptotic cell death in the retina occurs as a consequence of impaired turnover of mutant Rhodopsin via activation of stress-specific JNK and p38 mitogen-activated protein kinases. We proved that the absence of ERAD members significantly diminishes Rhodopsin-induced activation of JNK. We also showed that ERAD is important in the degradation and aggregation of both wild type and mutant Rhodopsin.

Methodology

Within my research project we used Drosophila as genetic tool. We also performed knock-down in mammalian cell lines (RNAi, sh-RNA), WB, SDS-PAGE/2D-Electrophoresis/MALDI-TOF-MS and fluorescence microscopy.
